Check spark plug cap. Drain puke tube. Do you wait for self-check to complete before starting? Do all warning lights go out after it finishes self-check (except oil)? Recently my DT300i, which ALWAYS starts in two revolutions, became reluctant like yours. Battery voltage while cranking was down to 9.5 volts. Anything below 10.5 volts means replace battery.  Like Stig said, your battery is probably the culprit. Yuasa the best.

I did all you mentioned before posting, only thing i did not measure voltage while starting. Must find voltmeter from someone before replacing my 4 year old battery. By the way, how many years have past since replacing factory battery and/or replacement battery?

Battery perhaps not as healthy as you think?
Could you pop for a new battery at this time?

Maybe, but after succesuful start I have tried starting a dozen times afterwards without a problem, it starts every time. Also I have put battery on charger and it was OK(2 of 4 blocks on digital charger display). Only thing that I can do is to buy a new battery and see, but it happens rarely(enough not to trust him anymore to go anywhere far from my city).

Have you tried starting the bike without the fuel cap?

Yes I did, and I did anti-flood procedure also, I suspect it has something with the check engine light while cranking but have no idea what that could be.(there is no error code on dashboard also)

Hello! I have bought my downtown 300(2014) in december and since then I had twice the same problem. The problem is when the engine is cranking and the yellow check engine light is lit on dashboard and the engine won`t start. And after a while(20min or so) it starts just like nothing`s happen. The spark plug is new, battery also good voltage, and I have no idea what is happening. Trouble is that I can`t go anywhere far now because I am afraid he will let me down and drain the battery while I`m trying to start the engine. Does anybody had similar problems? Thank you
